SOLU-KNEX® cleavable linker features a Val-Cit dipeptide. In this product, a DBCO-Dap(m-PEG4) moiety replaces the conventional maleimidocaproyl (MC) unit to enhance aqueous solubility. DBCO is a strained alkyne commonly used in copper-free click chemistry, specifically in the strain-promoted azide-alkyne cycloaddition (SPAAC) reaction. Additionally, m-PEG4 is attached to the commonly used self-immolative para-aminobenzyl (PAB) moiety to further increase aqueous solubility. The PNP (para-nitrophenyl) moiety serves as an activating leaving group, enabling efficient and selective conjugation of the payload to the linker.

Derived from established ADC linker architectures, SOLU-KNEX® ADC linkers integrate solubilizing moieties—such as PEG, amine-containing groups, or polysarcosine (PSAR), to enhance aqueous solubility. Solubility and physicochemical properties can be finely tuned through selection and combination of these groups, making the linkers highly adaptable for diverse ADC and related applications.
