Ac-Lys(Suc)-AMC is a modified fluorogenic peptide substrate widely used in enzymology to monitor deacylase and protease activities, especially those targeting lysine side-chain modifications. It consists of an N-terminal acetyl group, a Lysine residue bearing a succinyl (Suc) modification on its ε-amino group, and a C-terminal 7-Amino-4-methylcoumarin (AMC) moiety. The succinyl group mimics a biologically relevant post-translational modification (lysine succinylation), making the substrate particularly useful for studying enzymes such as sirtuins (e.g., SIRT5) that remove this modification. In a typical assay, the enzyme first removes the succinyl group, after which a secondary proteolytic step cleaves the peptide bond to release free AMC, generating a strong fluorescent signal. This two-step mechanism provides high specificity and sensitivity, enabling kinetic measurements, inhibitor screening, and studies of enzyme selectivity toward different lysine acylations.
