PC-6 is a specially designed fluorescent probe for detecting SARM1 activity, built by conjugating a styryl donor group to a pyridine acceptor to form a donor–π–acceptor system. It functions through SARM1’s base-exchange reaction, where PC-6 is converted into a positively charged product (PAD6) upon incorporation into NAD, leading to strong intramolecular charge transfer and a large fluorescence shift (from ~430 nm to ~520 nm). Among a series of related probes (PC1–9), PC-6 shows the largest fluorescence increase, high sensitivity (detecting as little as 48 ng/mL enzyme), and strong selectivity for SARM1 over other NADases, making it a powerful tool for monitoring SARM1 activation in biochemical assays.
