HaloTag is a self-labeling protein tag engineered from the haloalkane dehalogenase DhaA, originally isolated from Rhodococcus rhodochrous. Its active site binds chloroalkane (CA)substrates, forming an irreversible covalent bond. This stable linkage between the HaloTag protein and its ligand enables rapid and efficient isolation and purification of tagged proteins. CA-amide-PEG3-Biotin is a Biotin-HaloTag ligand that enables rapid labeling of Halo-tagged proteins and their efficient isolation via streptavidin pull-down. Biotin is a high-affinity ligand for streptavidin and avidin proteins. PEG3 linker enhances water solubility and provides spatial separation to minimize steric hindrance during binding interactions.
