HaloTag is a self-labeling protein tag engineered from the haloalkane dehalogenase DhaA, originally isolated from Rhodococcus rhodochrous. Its active site binds chloroalkane (CA)substrates, forming an irreversible covalent bond. This stable linkage between the HaloTag protein and its ligand enables rapid and efficient isolation and purification of tagged proteins. CA-Py-BODIPY (BODIPY 576/589) is a HaloTag Ligand containg Py-BODIPY dye and it forms a covalent bond with the HaloTag protein, ensuring stable labeling for long-term tracking. Py-BODIPY (576Ex/589Em) is widely used for its sharp absorption and emission peaks, high molar absorptivity, and excellent quantum yields. Py-BODIPY (BODIPY 576/589) is a small-molecule fluorescent and photoacoustic probe characterized by strong UV absorption, high photostability, and cell membrane permeability.
