HaloTag is a self-labeling protein tag engineered from the haloalkane dehalogenase DhaA, originally isolated from Rhodococcus rhodochrous. Its active site binds chloroalkane (CA)substrates, forming an irreversible covalent bond. This stable linkage between the HaloTag protein and its ligand enables rapid and efficient isolation and purification of tagged proteins. CA-Nor is a Halotag ligand containg norbornene moiety for click chemistry applications. Nor (norbornene) is a strained cyclic alkene that participates in inverse electron-demand Diels-Alder (iEDDA) reactions with tetrazines to form stable products. It is widely used in click chemistry for bioconjugation, fluorescent labeling, and drug delivery, thanks to its bioorthogonal nature and high reactivity due to its strained structure.
