HaloTag is a self-labeling protein tag engineered from the haloalkane dehalogenase DhaA, originally isolated from Rhodococcus rhodochrous. Its active site binds chloroalkane (CA)substrates, forming an irreversible covalent bond. This stable linkage between the HaloTag protein and its ligand enables rapid and efficient isolation and purification of tagged proteins. CA-PEG4-Lissamine Rhodamine B is a HaloTag Ligand containg Lissamine Rhodamine B dye. It forms a covalent bond with the HaloTag protein, ensuring stable labeling for long-term tracking. PEG4 linker enhances water solubility and provides spatial separation to minimize steric hindrance during binding interactions. Lissamine Rhodamine B (560Ex/581Em) is a sulfonated rhodamine derivative that fluoresces in the orange-red region. It offers high brightness, good photostability, and relatively pH-insensitive fluorescence, which are advantageous for consistent signal detection. The sulfonate groups enhance its water solubility and reduce nonspecific hydrophobic interactions.
