Fmoc-Val-Cit-PAB is a cleavable ADC linker featuring a Val–Cit dipeptide and a para-aminobenzyl (PAB) spacer. The Val–Cit sequence is selectively cleaved by cathepsin B in lysosomes, while the para-aminobenzyl (PAB) spacer supports self-immolative release of the attached drug, and the Fmoc protecting group can be removed to enable further functionalization or conjugation.
