Halo-DBF is a HaloTag-reactive dibromofluorescein photosensitizer designed for spatially controlled biomolecule labeling in living cells. Upon covalent binding to a HaloTag fusion protein, Halo-DBF localizes singlet-oxygen generation to a selected protein, organelle, or subcellular compartment. Visible-light activation oxidizes nearby guanosine residues in RNA, enabling capture by propargylamine and subsequent click-chemistry conjugation to biotin or fluorescent probes for enrichment, imaging, RT-qPCR, or RNA-seq analysis. This reagent supports proximity-based profiling of localized RNAs at nuclear, cytoplasmic, nucleolar, mitochondrial, and other defined cellular sites while preserving native cellular organization. Halo-DBF is well suited for RNA localization studies, transcriptome mapping, organelle-associated RNA analysis, and development of HaloTag-enabled chemical biology workflows.
