P18h is a HaloTag ligand incorporating the fluorogenic AggFluor P18 dye. AggFluor is derived from the synthetic GFP chromophore, 4-hydroxybenzylidene-imidazolinone (HBI), and belongs to the class of molecular rotor fluorophores widely used in biological imaging due to their sensitivity to environmental factors such as viscosity. When bound to properly folded proteins, AggFluors are quenched and remain non-fluorescent; however, protein misfolding or aggregation disrupts this quenching, resulting in turn-on fluorescence. AggFluor P1 exhibits fluorescence in both misfolded oligomers and insoluble aggregates but cannot differentiate between these species, as they can form morphologically similar granules. In contrast, AggFluor P18 selectively fluoresces in insoluble aggregates and shows high sensitivity to local viscosity. By combining P1 and P18, dual-color imaging enables the distinction between misfolded oligomers and insoluble aggregates in live cells. Beyond monitoring protein aggregation, AggFluor probes can also be applied to investigate biological processes involving spatial and temporal changes in local viscosity.
