HaXS8 is a synthetic chemical dimerizer designed to induce covalent and irreversible intracellular dimerization between proteins tagged with HaloTag and SNAP-tag. It’s a bifunctional molecule featuring a chloroalkane group (specific for HaloTag) and an O6-benzylguanine (BG) derivative (specific for SNAP-tag), connected by a flexible polyethylene glycol (PEG) linker. When introduced to cells, HaXS8 bridges HaloTag and SNAP-tag fusion proteins by reacting covalently with their respective active sites: the chloroalkane binds irreversibly to HaloTag’s modified dehalogenase pocket, while the BG moiety forms a stable thioether bond with SNAP-tag’s engineered alkyltransferase cysteine. This dual reactivity forces the tagged proteins into close proximity, creating a stable dimer without requiring additional catalysts.
