HaloTag is a self-labeling protein tag engineered from the haloalkane dehalogenase DhaA, originally isolated from Rhodococcus rhodochrous. Its active site binds chloroalkane (CA)substrates, forming an irreversible covalent bond. This stable linkage between the HaloTag protein and its ligand enables rapid and efficient isolation and purification of tagged proteins. CA-PEG1-Biotin is a cell-permeable Biotin-HaloTag ligand that enables rapid labeling of Halo-tagged proteins in live cells and their efficient isolation via streptavidin pull-down. It offers significant potential for studying fundamental cellular processes, such as genome duplication and chromatin maintenance.
