HaloTag is a self-labeling protein tag engineered from the haloalkane dehalogenase DhaA, originally isolated from Rhodococcus rhodochrous. Its active site binds chloroalkane (CA)substrates, forming an irreversible covalent bond. This stable linkage between the HaloTag protein and its ligand enables rapid and efficient isolation and purification of tagged proteins. In this product, the chloro atom in Biotin-PEG4-halo is replaced with a methyl group, preventing it from forming a covalent bond with the Halo-tag protein. This product serves as a neagtive control for Biotin-PEG4-Halotag Ligand. By replacing the chloro atom in the chloroalkane group with a methyl group, the resulting compound loses its ability to form this covalent bond. The methyl group is inert in this context and cannot undergo the nucleophilic substitution reaction that the chloroalkane relies on to bind the HaloTag protein. This modification effectively creates a structurally similar molecule that mimics the original ligand’s size, solubility, and biotin functionality but lacks reactivity with the HaloTag. As a negative control, it allows researchers to assess non-specific binding or background signals in experiments, distinguishing effects due to covalent attachment from those caused by other interactions (e.g., biotin-streptavidin binding or hydrophobic effects).
