CLIP-tag is a modified version of the SNAP-tag. It is also a self-labeling protein derived from human O6-alkylguanine-DNA-alkyltransferase. Unlike the SNAP-tag, which reacts with benzylguanine (BG)or chloropyrimidine (CP) derivatives, the CLIP-tag is engineered to specifically interact with benzylcytosine (BC)derivatives. This modification allows for distinct labeling applications, enabling researchers to use both SNAP-tag and CLIP-tag in the same experiment, providing flexibility in protein labeling and tracking. BC-Nor is a CLIP-tag substrate containg norbornene moiety for click chemistry applications. Nor (norbornene) is a strained cyclic alkene that participates in inverse electron-demand Diels-Alder (iEDDA) reactions with tetrazines to form stable products. It is widely used in click chemistry for bioconjugation, fluorescent labeling, and drug delivery, thanks to its bioorthogonal nature and high reactivity due to its strained structure.
