CLIP-tag is a modified version of the SNAP-tag. It is also a self-labeling protein derived from human O6-alkylguanine-DNA-alkyltransferase. Unlike the SNAP-tag, which reacts with benzylguanine (BG)or chloropyrimidine (CP) derivatives, the CLIP-tag is engineered to specifically interact with benzylcytosine (BC)derivatives. This modification allows for distinct labeling applications, enabling researchers to use both SNAP-tag and CLIP-tag in the same experiment, providing flexibility in protein labeling and tracking. BC-CPY is a CLIP-tag substrate containing the 6-CPY dye. It forms a covalent bond with the CLIP-tag protein, ensuring stable labeling for long-term tracking. CPY dye (short for carbopyronine, 606Ex/626Em) is a red-emitting fluorophore that emits strong, stable fluorescence in the red spectrum. This makes it ideal for high-resolution imaging techniques like STED (Stimulated Emission Depletion) and SIM (Structured Illumination Microscopy), as well as for tracking proteins in live-cell experiments. The photostability and brightness of CPY make it a valuable tool for detailed imaging of cellular dynamics.
