CLIP-tag is a modified version of the SNAP-tag. It is also a self-labeling protein derived from human O6-alkylguanine-DNA-alkyltransferase. Unlike the SNAP-tag, which reacts with benzylguanine (BG)or chloropyrimidine (CP) derivatives, the CLIP-tag is engineered to specifically interact with benzylcytosine (BC)derivatives. This modification allows for distinct labeling applications, enabling researchers to use both SNAP-tag and CLIP-tag in the same experiment, providing flexibility in protein labeling and tracking. BC-TCO is a CLIP-tag substrate containg TCO moiety for click chemistry chemistry applications. TCO (trans-cyclooctene) is a strained alkene used in click chemistry for bioorthogonal reactions like iEDDA (inverse electron-demand Diels-Alder).
