HaloTag is a self-labeling protein tag engineered from the haloalkane dehalogenase DhaA, originally isolated from Rhodococcus rhodochrous. Its active site binds chloroalkane (CA)substrates, forming an irreversible covalent bond. This stable linkage between the HaloTag protein and its ligand enables rapid and efficient isolation and purification of tagged proteins. CA-BODIPY is a HaloTag Ligand containg BODIPY dye and it forms a covalent bond with the HaloTag protein, ensuring stable labeling for long-term tracking. BODIPY (boron-dipyrromethene, 502Ex/511Em) is widely used for its sharp absorption and emission peaks, high molar absorptivity, and excellent quantum yields. The rigid, planar structure around the boron-dipyrromethene core imparts exceptional photostability, minimal environmental sensitivity, and narrow spectral bands, making them ideal for multiplexed fluorescence applications.
