HaloTag is a self-labeling protein tag engineered from the haloalkane dehalogenase DhaA, originally isolated from Rhodococcus rhodochrous. Its active site binds chloroalkane (CA)substrates, forming an irreversible covalent bond. This stable linkage between the HaloTag protein and its ligand enables rapid and efficient isolation and purification of tagged proteins. CA-PEG4-amide-PEG2-Bition is a Biotin-HaloTag ligand that enables rapid labeling of Halo-tagged proteins and their efficient isolation via streptavidin pull-down. Biotin is a high-affinity ligand for streptavidin and avidin proteins. PEG linkers enhance water solubility and provide spatial separation to minimize steric hindrance during binding interactions.
