HaloTag is a self-labeling protein tag engineered from the haloalkane dehalogenase DhaA, originally isolated from Rhodococcus rhodochrous. Its active site binds chloroalkane (CA)substrates, forming an irreversible covalent bond. This stable linkage between the HaloTag protein and its ligand enables rapid and efficient isolation and purification of tagged proteins. CA-SS-Alexa488 is a cleavable, membrane-impermeable Halotag ligand. It features a disulfide bond, allowing the non-internalized probe to be released from the cell surface using the reducing agent sodium 2-mercaptoethanesulfonate (MESNA). In vitro, CA-SS–Alexa488 fully labels recombinantly expressed Halo-tag proteins, with the fluorophore removable by treatment with 100 mM sodium 2-mercaptoethanesulfonate (MESNA). In live HEK 293T cells, CA-SS–Alexa488 efficiently labels surface-expressed Halo-mGluR2, and a brief treatment with 100 mM MESNA results in a near-complete loss of fluorescence. Alexa Fluor 488 (494Ex/517Em) is a popular green fluorescent dye known for its brightness, stability, and versatility in various biological and biochemical applications.
