HaloTag is a self-labeling protein tag engineered from the haloalkane dehalogenase DhaA, originally isolated from Rhodococcus rhodochrous. Its active site binds chloroalkane (CA)substrates, forming an irreversible covalent bond. This stable linkage between the HaloTag protein and its ligand enables rapid and efficient isolation and purification of tagged proteins. CA-SulfoCy3 is a HaloTag ligand with SulfoCyanine3 dye. SulfoCy3(Sulfo-Cyanine3, 548Ex/562Em) is a sulfonated derivative of the Cy3 dye, with the addition of sulfonic acid groups that enhance its solubility in aqueous environments. This dye hass bright fluorescence, high extinction coefficient, and photostability, which allow for sensitive detection in various imaging and analytical techniques. Its fluorescence is relatively pH-insensitive, and it exhibits reduced dye aggregation compared to non-sulfonated cyanine dyes, improving its performance in labeling applications.
