HaloTag is a self-labeling protein tag engineered from the haloalkane dehalogenase DhaA, originally isolated from Rhodococcus rhodochrous. Its active site binds chloroalkane (CA)substrates, forming an irreversible covalent bond. This stable linkage between the HaloTag protein and its ligand enables rapid and efficient isolation and purification of tagged proteins. Sulfo-CA-SiR is a sulfonated HaloTag substrate featuring a sulfonate group on the amide bond linking the chloroalkane moiety to the SiR dye. SiR (SiR-6-COOH, 652Ex/674Em) is a far-red fluorescent dye that is bright, photostable, and cell-permeable, and it is fully compatible with standard Cy5 filter sets.
