HaloTag is a self-labeling protein tag engineered from the haloalkane dehalogenase DhaA, originally isolated from Rhodococcus rhodochrous. Its active site binds chloroalkane (CA)substrates, forming an irreversible covalent bond. This stable linkage between the HaloTag protein and its ligand enables rapid and efficient isolation and purification of tagged proteins. CA-PhN3 is a HaloTag ligand containing an azide functional group, which readily reacts with an alkyne via click chemistry. In click chemistry, azides are commonly used in azide-alkyne cycloaddition reactions, such as the copper-catalyzed azide-alkyne cycloaddition (CuAAC) and the strain-promoted alkyne-azide cycloaddition (SPAAC). These reactions are efficient and selective, forming a triazole ring and often used in drug development, molecular biology, and bioconjugation.
