HaloTag is a self-labeling protein tag engineered from the haloalkane dehalogenase DhaA, originally isolated from Rhodococcus rhodochrous. Its active site binds chloroalkane (CA)substrates, forming an irreversible covalent bond. This stable linkage between the HaloTag protein and its ligand enables rapid and efficient isolation and purification of tagged proteins. CA-SS-TMR is a cleavable HaloTag ligand featuring a disulfide bond that can be cleaved using a reducing agent, such as sodium 2-mercaptoethanesulfonate (MESNA). TMR dye (555Ex/585Em) provides strong red fluorescence, enabling researchers to visualize and track the protein of interest in live cells or tissue using fluorescence microscopy or imaging systems.
