SNAP-tag is a self-labeling protein derived from human O6-alkylguanine-DNA-alkyltransferase. At 20 kDa, SNAP-tag is smaller than HaloTag (33 kDa), potentially reducing interference with the tagged protein’s function. SNAP-tag reacts with covalently with benzylguanine (BG)or chloropyrimidine (CP) derivatives, forming a stable thioether bonds that ensures durability for long-term experiments or harsh conditions. SNAP-tag can be used alongside other tags like CLIP-tag for multicolor labeling. BG-SS-SulfoCy3 is a membrane-impermeable probe that covalently binds to the SNAP-tag, emitting in the red spectrum. It features a disulfide bond, allowing the non-internalized probe to be released from the cell surface using the reducing agent sodium 2-mercaptoethanesulfonate (MESNA). Together, BG-SS-SulfoCy3 and BG-SS-SulfoCy5 have been utilized for the simultaneous visualization of multiple internalized cell surface proteins in two colors, exemplified by studies involving G protein-coupled receptors. SulfoCy3(Sulfo-Cyanine3, 548Ex/562Em) is a sulfonated derivative of the Cy3 dye, with the addition of sulfonic acid groups that enhance its solubility in aqueous environments. This dye hass bright fluorescence, high extinction coefficient, and photostability, which allow for sensitive detection in various imaging and analytical techniques. Its fluorescence is relatively pH-insensitive, and it exhibits reduced dye aggregation compared to non-sulfonated cyanine dyes, improving its performance in labeling applications.
