HaloTag is a self-labeling protein tag engineered from the haloalkane dehalogenase DhaA, originally isolated from Rhodococcus rhodochrous. Its active site binds chloroalkane (CA)substrates, forming an irreversible covalent bond. This stable linkage between the HaloTag protein and its ligand enables rapid and efficient isolation and purification of tagged proteins. TMR-SHTL is a sulfonated HaloTag substrate featuring a sulfonate group on the amide bond linking the chloroalkane moiety to the TMR dye. In a study using TMR-d12-SHTL, the introduction of the charged sulfonate group minimally altered the excitation and emission profiles compared to the non-sulfonated TMR-d12 (HTL: ?Ex/Em = 553/577 nm; SHTL: ?Ex/Em = 554/579 nm). Additionally, it showed no significant change in labeling kinetics, exhibited longer fluorescence lifetimes than its non-sulfonated counterparts, and demonstrated selective surface labeling.
