HaloTag is a self-labeling protein tag engineered from the haloalkane dehalogenase DhaA, originally isolated from Rhodococcus rhodochrous. Its active site binds chloroalkane (CA)substrates, forming an irreversible covalent bond. This stable linkage between the HaloTag protein and its ligand enables rapid and efficient isolation and purification of tagged proteins. CA-SS-Lissamine Rhodamine B is a cleavable HaloTag ligand featuring a disulfide bond that can be cleaved using a reducing agent, such as sodium 2-mercaptoethanesulfonate (MESNA). Lissamine Rhodamine B (560Ex/581Em) is a sulfonated rhodamine derivative that fluoresces in the orange-red region. It offers high brightness, good photostability, and relatively pH-insensitive fluorescence, which are advantageous for consistent signal detection. The sulfonate groups enhance its water solubility and reduce nonspecific hydrophobic interactions.
