Cy5 Labeled Thalidomide

Cy5-labeled thalidomide is a far-red fluorescent probe for studying cereblon (CRBN), a substrate receptor in the CRL4–CRBN E3 ubiquitin ligase complex. The Cy5-conjugated thalidomide probe exhibits a reported binding affinity ($K_d$) of 121.6 nM and supports competitive fluorescence polarization assays for evaluating CRBN-binding compounds. Reported IC₅₀ values were 404.6 nM for thalidomide, 296.9 nM for lenalidomide, and 264.8 nM for pomalidomide. Thalidomide–Cyanine 5 is suitable for TR-FRET, with absorption and emission maxima of approximately 650 and 665 nm, respectively. These properties make fluorescent thalidomide probes useful tools for CRBN ligand screening and targeted protein degradation research.
