MaP555-Halo is a highly cell-permeable and fluorogenic probe for live-cell imaging. MaP555 is a small-molecule fluorogenic probe with excellent cell permeability. It is converted from 6-carboxytetramethylrhodamine (6-TAMRA) and has similar spectroscopic properties of 6-TAMRA. MaP555 has an acyl sulfamide structure which favors the spirolactam form and thus has excellent cell permeability. When the probe binds to its cellular targets, MaP555 mainly exists in the fluorescent zwitterion form. The good spectroscopic properties, high cell permeability and outstanding fluorogenicity of MaP555 make it very useful for wash-free, multicolour, live-cell nanoscopy.
